Antigen/Purification: ExpandCollapse

The antigen is a phosphopeptide corresponding to amino acid residues surrounding the phospho-Ser513 of Munc-18.

The antibody is prepared from rabbit serum by affinity purification via sequential chromatography on phospho- and dephospho-peptide affinity columns.

Biological Significance: ExpandCollapse

Munc-18 (mammalian homologue of Unc-18) is a protein that is thought to be involved in regulating exocytosis due, at least in part, to it ability to bind syntaxin (Ciufo et al., 2005). Munc18-1 is a neuron-specific member of the Sec1/Munc18 protein family that binds to syntaxin1A and is thought to stabilize the complex (Liu et al., 2004). The function of Unc-18 is thought to be regulated by PKC phosphorylation of Ser515 in C. Elegans (Sassa et al., 1996). Ser513 is the mammalian homologue of Ser515 in C. Elegans.

100 µl in 10 mM HEPES (pH 7.5), 150 mM NaCl, 100 µg BSA per ml and 50% glycerol. Adequate amount of material to conduct 10-mini Western Blots.

For long term storage –20° C is recommended. Stable at –20° C for at least 1 year.

Munc-18 ser513 Antibody
Western blot of rat cortical lysate showing specific immunolabeling of the ~65 kDa Munc-18 protein phosphorylated at Ser513 in the first lane (-). Phosphospecificity is shown in the second lane (+) where immunolabeling is blocked by preadsorption of the phosphopeptide used as the antigen, but not by the corresponding non-phosphopeptide (not shown).
Immunostaining of 14 DIV rat cortical neurons showing specific labeling of Munc-18 phosphorylated at Ser513 (red) and PSD95 (green).
